Notch通路与支气管哮喘相关的研究进展

近年来大量研究表明Notch通路在支气管哮喘(哮喘)的发生、发展中有十分重要的作用。Notch通路调控肺组织的发生发育,可以决定细胞的分化方向、调控肺泡和肺血管的发育;Notch通路参与T细胞调节,通过改变Th1/Th2平衡,影响Th17、Treg、树突状细胞表达等途径,导致哮喘的发生、发展;Notch通路也通过改变各种炎性细胞如淋巴细胞、嗜酸粒细胞的浸润,促进气道上皮杯状细胞化生与气道黏液分泌等途径,参与了哮喘气道重塑的病理改变。同时针对Notch通路上的靶点,对于哮喘治疗方法、治疗药物的开发也取得了许多新成果,包括γ-分泌酶抑制剂、干细胞、树突状细胞以及我国传统中医药等在不同模型中已被证明对哮喘有效。但对于Notch通路,其各种调控机制并未完全阐明,需要更加深入的研究;其应用于临床治疗的价值仍有待进一步发掘。本文就Notch信号通路与哮喘的相关研究,进行综述如下。“,”In recent years, a large number of studies have shown that Notch pathway plays an important role in the occurrence and development of bronchial asthma(asthma). Notch pathway regulates the development of lung tissue, which determines the direction of cell differentiation, regulate the development of alveoli and pulmonary vessels.Notch pathway participates in the regulation of T cells, which leads to asthma by changing the balance of Th1 / Th2, affecting the expression of Th17, Treg and dendritic cells.Notch pathway also participates in the pathological changes of airway remodeling by changing the expression of inflammatory cells such as lymphocytes and eosinophils, promoting the goblet cell metaplasia of airway epithelium and airway mucus secretion.At the same time, aiming Notch pathway, many new achievements have been made in the research of asthma treatment methods and the development of therapeutic drugs, including γ-secretase inhibitor, stem cells, dendritic cells and Chinese traditional medicine, are proved to be effective in treating asthma on different models.However, for Notch pathway, its various regulatory mechanisms are not fully elucidated and need more in-depth studies.The value of its application in clinical treatment remains to be further explored.In this paper, related researches between Notch pathway and asthma are summarized as follows.
