亚低温对凝血酶所致大鼠脑水肿及神经细胞凋亡的影响

目的 探讨亚低温对凝血酶所致大鼠脑组织水肿及神经细胞凋亡的影响。方法 通过立体定位向大鼠颅内(基底节区)注射凝血酶,建立凝血酶所致的脑组织损伤大鼠模型。用干、湿重法测定脑组织水肿程度,用碘化丙啶(PI)染色流式细胞仪及透射电镜检测细胞凋亡百分率,分别观察了常温凝血酶组、常温对照组、亚低温凝血酶组和亚低温对照组大鼠脑组织水肿和神经细胞凋亡百分率。结果 与常温凝血酶组相比,亚低温凝血酶组大鼠脑组织水肿明显减轻(P<0.05),凋亡细胞百分率也明显降低(P<0.05)。结论 亚低温能减轻凝血酶所致脑组织水肿和减少神经细胞凋亡。 Objective To investigate the effects of mild hypothermia on thrombin-induced edema and neuronal apoptosis in rat brain. Methods Thrombin was injected intracranially (basal ganglia) into rats by stereotaxic method to establish rat model of thrombin-induced brain injury. The degree of brain edema was determined by dry and wet weight method. The percentage of apoptosis was detected by PI staining and transmission electron microscopy. The changes of apoptosis in normal temperature thrombin group, normal temperature control group, mild hypothermia thrombin Group and mild hypothermia control group rats brain edema and nerve cell apoptosis percentage. Results Compared with normal thrombin group, the edema of brain tissue in mild hypothermic thrombin group was significantly reduced (P <0.05) and the percentage of apoptotic cells was also significantly decreased (P <0.05). Conclusion Mild hypothermia can reduce thrombin-induced edema in brain tissue and reduce apoptosis of nerve cells.
