目的 :观察低氧时大鼠肺动脉平滑肌细胞 (PASMC)受血管舒张因子一氧化碳的作用。方法 :随机分为常氧组、低氧组 ( 1%O2 +5 %CO2 +94%N2 )、一氧化碳组 ( 3%CO +5 %CO2 +92 %N2 ) ,采用流式细胞仪、增殖细胞核抗原 (PCNA)、[3H]-胸腺嘧啶 ( [3 H]-TdR)摄取量 ,检测PASMC增生的情况。结果 :( 1)流式细胞仪检测低氧组PASMCG2 M期细胞比例明显增多(P <0 .0 1) ,G0 /G1 期细胞比例明显减少 (P <0 .0 1) ,CO组与常氧组的PASMC各期细胞数差异不显著 (P >0 .0 5 )。 ( 2 )PCNA免疫组化染色 ,低氧组PASMC细胞核阳性反应颗粒明显增加 ,而CO组为弱阳性。 ( 3) [3 H]-TdR的摄取量 ,低氧组可明显促进PASMC摄取 [3H]-TdR (P <0 .0 5 ) ,CO组与常氧组比没有明显差异 (P >0 .0 5 )。结论 :低氧可以促进PASMC的增殖 ,而CO可以部分抑制低氧对PASMC的作用
Objective: To observe the effect of hypoxia on pulmonary artery smooth muscle cells (PASMC) by the vasodilator carbon monoxide. Methods: The rats were randomly divided into normoxia group, hypoxia group (1% O2 +5% CO2 + 94% N2) and carbon monoxide group (3% CO +5% CO2 +92% N2). Flow cytometry Antigen (PCNA), [3H] -thymidine ([3H] -TdR) uptake were measured to detect the proliferation of PASMC. Results: (1) The percentage of M phase cells in PASMCG was significantly increased in hypoxia group (P <0.01) by flow cytometry, the proportion of cells in G0 / G1 phase was significantly decreased (P <0.01) There was no significant difference in the number of PASMC in oxygen group (P> 0.05). (2) PCNA immunohistochemical staining, PASMC nuclear positive particles in hypoxia group increased significantly, while CO group was weakly positive. (3) The intake of [3 H] -TdR, hypoxia group could obviously promote the uptake of [3H] -TdR in PASMC (P <0.05). There was no significant difference between CO group and normoxia group (P> 0. 0 5). Conclusion: Hypoxia can promote the proliferation of PASMC, while CO can partially inhibit the effect of hypoxia on PASMC
