目的初步探讨聚腺苷二磷酸核糖聚合酶(PARP)与大肠癌侵袭转移的关系及其可能机制。方法采用SP免疫组化法检测PAR(聚腺苷二磷酸核糖,系PARP产物)、P-selectin及ICAM-1的表达;并应用免疫荧光双标法检测PAR与P-selectin、ICAM-1的共表达。结果大肠癌组织内PAR、P-selectin及ICAM-1的表达均明显高于对照肠黏膜(P<0.05);3种蛋白的表达仅与大肠癌转移有关(P<0.05),而与其他临床病理因素无关;PAR与P-selectin和ICAM-1的表达均呈正相关关系(P<0.05)。结论大肠癌组织内PARP活性增强,并可能通过上调P-selectin和ICAM-1的表达而有利于大肠癌侵袭转移,有望成为判断大肠癌转移的参考指标之一。
Objective To investigate the relationship between poly (ADP-ribose) polymerase (PARP) and invasion and metastasis of colorectal carcinoma and its possible mechanism. Methods The expression of PAR (poly-ADP ribosomal PARP product), P-selectin and ICAM-1 were detected by SP immunohistochemistry. The expressions of PAR and P-selectin, ICAM-1 Common expression. Results The expressions of PAR, P-selectin and ICAM-1 in colorectal cancer tissues were significantly higher than those in control intestinal mucosa (P <0.05). The expressions of three proteins were only correlated with the metastasis of colorectal carcinoma (P <0.05) There was a positive correlation between PAR and P-selectin and ICAM-1 (P <0.05). Conclusion The activity of PARP in colorectal cancer is increased, which may be beneficial to the invasion and metastasis of colorectal cancer by up-regulating the expression of P-selectin and ICAM-1. It may be one of the reference indexes for judging the metastasis of colorectal cancer.
