目的探讨植物雌激素α-ZAL影响oxLDL诱导的人脐静脉内皮细胞分泌NO、ET-1的机制。方法采用RTPCR方法对eNOS、iNOS、ET-1mRNA的表达进行分析;采用WesternBlotting方法对eNOS、iNOS蛋白表达进行分析;同时测定内皮细胞上清液中NO、ET-1分泌的变化。结果植物雌激素α-ZAL增加oxLDL诱导内皮细胞eNOSmRNA、蛋白的表达和NO的分泌,降低ET-1mRNA的表达和蛋白的分泌。结论植物雌激素α-ZAL通过上调oxLDL诱导内皮细胞eNOSmRNA、蛋白的表达和NO的分泌,下调ET1mRNA的表达和蛋白的分泌起着保护血管内皮的作用。
Objective To investigate the mechanism of phytoestrogen α-ZAL in oxLDL-induced secretion of NO and ET-1 in human umbilical vein endothelial cells. Methods The expressions of eNOS, iNOS and ET-1 mRNA were analyzed by RTPCR. The expressions of eNOS and iNOS were analyzed by Western Blotting. The changes of NO and ET-1 secretion in the supernatant of endothelial cells were also assayed. Results Phytoestrogen α-ZAL increased oxLDL-induced eNOS mRNA and protein expression and NO secretion in endothelial cells, decreased ET-1 mRNA expression and protein secretion. Conclusion Phytoestrogen α-ZAL plays an important role in the protection of vascular endothelium by up-regulating the expression of eNOS mRNA and protein, the secretion of NO and the expression of ET1mRNA in endothelial cells.
