目的探讨人IL-16重组腺病毒对Hela细胞增殖和凋亡的影响。方法用IL-16重组腺病毒感染Hela细胞后,不同时间收获细胞。MTT法检测细胞增殖,藻红蛋白标记的annexin V和7-氨基放线菌素D联合染色(annexin V-PE/7-AAD)结合流式细胞术检测感染后Hela细胞的凋亡、Western blot法检测Hela细胞中HPV病毒相关蛋白p16、E7以及凋亡相关蛋白Bax、Bcl-2的表达。结果 MTT法结果证明IL-16腺病毒感染的Hela细胞增殖受到明显抑制;流式细胞术检测结果表明细胞凋亡增加,Western blot法证实HPV病毒相关蛋白P16、E7表达降低,Bax蛋白表达量增强,而Bcl-2蛋白表达量减弱,且呈现一定程度的时间依赖性。结论人IL-16重组腺病毒可通过下调Bcl-2蛋白的表达和上调Bax蛋白的表达诱导Hela细胞凋亡。
Objective To investigate the effects of human IL-16 recombinant adenovirus on proliferation and apoptosis of Hela cells. Methods Hela cells were infected with IL-16 recombinant adenovirus and cells were harvested at different times. The apoptosis of Hela cells was detected by MTT assay, the apoptosis of Hela cells was detected by annexin V and annexin V-PE / 7-AAD combined with flow cytometry, Western blot Method to detect the expression of HPV16, E7 and Bax, Bcl-2 in Hela cells. Results The results of MTT assay showed that the proliferation of Hela cells infected with IL-16 was significantly inhibited. The results of flow cytometry showed that the apoptosis of Hela cells was increased. Western blot showed that the expression of P16 and E7 was decreased and the expression of Bax protein was enhanced , While the expression of Bcl-2 protein decreased, and showed a certain degree of time-dependent. Conclusion Human IL-16 recombinant adenovirus can induce Hela cell apoptosis by down-regulating Bcl-2 protein expression and up-regulating Bax protein expression.
