目的:探讨3种共刺激分子B7-H1、B7-H3和B7-H4在人类血液系统恶性肿瘤细胞株中的表达及分布特征。方法:分别采用RT-PCR、q PCR、Western blot和流式细胞术检测13株血液肿瘤细胞中B7-H1、B7-H3和B7-H4的表达及亚细胞定位情况,设12例志愿者外周血单个核细胞(PB MNC)为对照。结果:3种共刺激分子的mRNA在12例志愿者PB MNC和13株血液肿瘤细胞中广泛表达,其中B7-H4表达水平相对偏低;3种共刺激分子分别在Maver、Z138和HL-60中表达最高,B7-H3和B7-H4在CZ1中不表达。3种共刺激分子的胞核及胞浆蛋白仅在恶性血液肿瘤细胞中异常高表达,分别在U937、Z138和Raji细胞中表达最高,B7-H3和B7-H4在CZ1细胞中不表达。在同一肿瘤来源的细胞株中,3种共刺激分子的mRNA和胞核及胞浆蛋白表达均存在差异,且差异不全一致。B7-H3膜蛋白在U937、M aver和Z138中表达丰度较高;B7-H1和B7-H4的膜蛋白在13株细胞中呈不表达或低表达。结论:共刺激分子B7-H1、B7-H3和B7-H4在mRNA水平均广泛表达,但蛋白水平仅在血液肿瘤细胞中异常高表达,且亚细胞定位多在胞核及胞浆,膜蛋白水平普遍低表达或不表达。同一肿瘤来源的细胞株中3种共刺激分子的表达可存在差异。
Objective: To investigate the expression and distribution of three co-stimulatory molecules B7-H1, B7-H3 and B7-H4 in human hematological malignancies. Methods: The expression and subcellular localization of B7-H1, B7-H3 and B7-H4 in 13 hematological malignancies were detected by RT-PCR, qPCR, Western blot and flow cytometry respectively. Blood mononuclear cells (PB MNC) served as controls. Results: The mRNA of the three costimulatory molecules was widely expressed in 12 volunteers PB MNC and 13 hematological malignancies, and the expression of B7-H4 was relatively low. The three costimulatory molecules were expressed in Maver, Z138 and HL-60 The highest expression, B7-H3 and B7-H4 is not expressed in CZ1. The nuclear and cytoplasmic proteins of the three costimulatory molecules were only highly expressed in malignant hematological tumor cells, and were the highest expressed in U937, Z138 and Raji cells, respectively. However, B7-H3 and B7-H4 were not expressed in CZ1 cells. In the same tumor-derived cell lines, there were differences in mRNA and nuclear and cytoplasmic protein expression of the three co-stimulatory molecules, and the differences were not identical. The B7-H3 membrane protein was highly expressed in U937, M aver and Z138. The membrane proteins of B7-H1 and B7-H4 were not expressed or down-regulated in 13 cells. CONCLUSIONS: The co-stimulatory molecules B7-H1, B7-H3 and B7-H4 are widely expressed at mRNA level, but the protein levels are only highly expressed in hematological malignancies, and subcellular localization is mostly in the nucleus and cytoplasm, membrane proteins The level is generally low or no expression. There may be differences in the expression of three co-stimulatory molecules in the same tumor-derived cell line.
