目的建立测定人血浆中氟罗沙星浓度的高效液相色谱法。方法血浆样品用三氟醋酸沉淀后离心,取上清液直接进样。色谱柱为ZORBAXC18柱(150mm×4.6mm,5μm),流动相为乙腈水(0.05%磷酸和0.05%三乙胺)(20∶80),流速为1.0mL·min-1,检测波长为286nm,柱温为室温。结果本方法线性范围49.875~7980ng·mL-1,r=0.99954,最小检出浓度为49.875ng·mL-1。绝对回收率均在70%以上,相对回收率在98%~102%,日内、日间RSD均<15%。结论本方法简便、灵敏、准确,可用于氟罗沙星血药浓度检测和药物动力学研究。 Objective To establish a HPLC method for the determination of fleroxacin in human plasma. Methods Plasma samples were centrifuged with trifluoroacetic acid and the supernatant was injected directly. The column was a ZORBAXC18 column (150 mm × 4.6 mm, 5 μm) with a mobile phase of acetonitrile (0.05% phosphoric acid and 0.05% triethylamine) (20:80) at a flow rate of 1.0 mL · min -1 with a detection wavelength of 286 nm. The column temperature is room temperature. Results The linear range of this method was 49.875 ~ 7980 ng · mL-1, r = 0.99954, the minimum detectable concentration was 49.875 ng · mL-1. The absolute recovery rates were above 70%, the relative recovery rate was 98% ~ 102%, day and day RSD were <15%. Conclusion The method is simple, sensitive and accurate and can be applied to the determination of fleroxacin and its pharmacokinetics.
