目的探讨柴陈片抗抑郁作用及其作用机理。方法对小鼠进行强迫游泳、悬尾、高架十字迷宫、爬梯、自主活动测定实验;结合慢性不可预见轻度应激抑郁大鼠模型,研究柴陈片的抗抑郁、抗焦虑作用。结果与空白对照组比较,柴陈片能缩短小鼠强迫游泳及悬尾的不动时间(P<0.01),增高小鼠开臂次数(OE)和开臂时间(OT)的百分比,减少站立次数(P<0.05,P<0.01)。与模型组比较,柴陈片组体质量增长明显,大鼠垂直运动次数和水平运动次数均明显增加,糖水偏爱度明显增高,血清中SOD、GSH-Px活力增加,MDA含量下降,差异均有统计学意义(P<0.05~0.01);各剂量组的柴陈片均不影响小鼠的自主活动。结论柴陈片能改善行为绝望模型小鼠的抑郁行为,能改善焦虑模型小鼠的矛盾心理和焦虑行为,改善慢性应激大鼠的抑郁症状,不具有中枢兴奋性。改善机体脂质过氧化状态可能其作用机制之一。
Objective To investigate the antidepressant effect and mechanism of Cisan tablets. Methods The mice were subjected to forced swimming test, tail suspension test, elevated plus maze test, stair climbing test and voluntary activity test. Combined with the chronic unpredictable mild stress depression rat model, the anti-depression and anti-anxiety effects of Cisan tablets were studied. Results Compared with the blank control group, Cisan tablets could shorten the forced swimming and tail-suspending immobility time (P <0.01), increase the percentage of open arms (OE) and open-arm time (OT) Frequency (P <0.05, P <0.01). Compared with the model group, the body weight of Chaihualian tablet group increased obviously, the number of vertical movement and horizontal movement of rats increased significantly, the degree of preference of syrup increased significantly, the activity of SOD and GSH-Px in serum increased, the content of MDA decreased Statistical significance (P <0.05 ~ 0.01). The Cicada films of each dose group did not affect the autonomic activity of mice. Conclusion Cisan tablets can improve the depression behavior of mice with behavioral desperation, improve the contradictory psychology and anxiety behavior of anxiety model mice, and improve the depressive symptoms of chronic stress rats without central excitability. Improve the body’s lipid peroxidation may be one of the mechanisms of action.
