核糖核酸的制备和分析受多种因素的影响.因此,在特定条件下制备高纯度和完整性好的RNA的方法研究至关重要.本文以大白鼠肝为材料,研究RNA的制备方法,建立了有效的RNA制备方法.用紫外分光光度计和变性电泳分析所制备的RNA,发现其纯度A260A280=1.7~2.0,28SrRNA与18SrRNA的带宽比为21.本方法具有节约节时,重复性好,产量较高,无DNA污染等优点.这为cDNA合成,基因表达和调控等研究奠定了基础.
The preparation and analysis of ribonucleic acid is influenced by many factors. Therefore, the study on the method of preparation of RNA with high purity and integrity under certain conditions is of great importance. In this paper, the rat liver as a material to study the preparation of RNA, established an effective preparation of RNA. The prepared RNA was analyzed by ultraviolet spectrophotometer and denaturing electrophoresis. The purity of the RNA was found to be A260A280 = 1.7 ~ 2.0. The bandwidth ratio of 28SrRNA to 18SrRNA was 21. The method has the advantages of saving time, good repeatability, high yield, no DNA pollution and the like. This laid the foundation for the study of cDNA synthesis, gene expression and regulation.
