本实验采用了PAP和胶体金免疫电镜双标记技术,对下丘脑室旁核神经元內进行标记,证实了室旁核内有血管加压素与生长抑素共存的神经元.为了加强抗原的固定效果,提高免疫反应的敏感性,我们采用了丙烯醛固定的方法,对比了浸泡固定和灌流固定的效果,在电镜水平证实了丙烯醛浸泡固定同灌流固定一样,均能获得良好的神经组织的超微结构和阳性反应。同时,我们分别采用了包埋前PAP与胶体金双标记和包埋前PAP结合包埋后胶体金双标记两种方法。结果表明,包埋后胶体金染色比包埋前较易发现阳性标记。
In this study, PAP and colloidal gold immunoelectron microscopy double labeling technique was used to label neurons in the paraventricular hypothalamic nucleus, which confirmed the existence of neurons coexisting with vasopressin and somatostatin in paraventricular nucleus.In order to strengthen the antigen Fixed effects and improve the sensitivity of the immune response, we used the acrolein fixation method, compared the effect of immersion fixation and perfusion fixation, the level of electron microscopy confirmed that acrolein immersion fixed with perfusion fixation, can get good nerve tissue Ultrastructure and positive reaction. At the same time, we used two methods, namely the double labeling of PAP and colloidal gold before embedding and the double labeling of colloidal gold after PAP embedding. The results show that, after embedding colloidal gold staining easier to find positive markers than before embedding.
