无籽是香水柠檬的重要特征,自交不亲和是香水柠檬无籽的主要原因,为了深入了解香水柠檬自交不亲和的分子机理。本研究在通过De novo技术获得F-box基因片段的基础上,以香水柠檬嫩叶为材料,利用RT-PCR技术成功地克隆出长度为1 357 bp的F-box基因,命名为LFB2。生物信息学分析发现该基因编码446个氨基酸,氨基酸的C端具有两个WD结构域,属于FBXW蛋白,其蛋白分子量为111.62 kD,等电点:8.97。利用qRT-PCR分析LFB2基因在花器官不同组织的表达情况,发现该基因在雄蕊中的表达量最高,是其它花器官的十倍左右;在逆境胁迫下,该基因的表达量也有明显的变化,以上研究表明LFB2基因对花粉的发育和逆境胁迫中有一定的作用。
Seedless is an important feature of lemon perfume, self-incompatibility is the main reason for lemon seedless perfume, in order to understand the molecular mechanism of lemon self-incompatibility. In this study, we obtained the F-box gene fragment by De novo technology, and successfully cloned the F-box gene with the length of 1 357 bp by RT-PCR and named LFB2. Bioinformatics analysis revealed that the gene encodes 446 amino acids. The amino acid has two WD domains at the C-terminus, belonging to the FBXW protein with a molecular weight of 111.62 kD and an isoelectric point of 8.97. The expression of LFB2 gene in different organs of floral organs was analyzed by qRT-PCR. The results showed that the expression level of LFB2 gene in stamens was about ten times that of other flower organs. Under the stress of stress, the expression level of LFB2 gene also changed significantly The above studies show that LFB2 gene has a certain effect on pollen development and stress.
