目的:探讨消瘀降脂胶囊对载脂蛋白E基因敲除(ApoEn -/-)小鼠血脂、颈动脉粥样硬化程度及斑块的影响。n 方法:ApoEn -/-小鼠采用高脂饲料饲养,建立颈动脉粥样硬化模型。将ApoEn -/-小鼠按体重分为模型组、阿托伐他汀组及消瘀降脂胶囊低、高剂量组,每组22只。以12只C57BL/6Cnc小鼠为对照组,采用普通饲料喂养。阿托伐他汀组灌胃阿托伐他汀混悬液1.3 mg/kg,消瘀降脂胶囊低、高剂量组灌胃消瘀降脂胶囊混悬液325、975 mg/kg,对照组和模型组灌胃等体积的蒸馏水。按0.1 ml/10 g小鼠体重灌胃,1次/d,每周记录小鼠体重。连续灌胃12周后,检测小鼠血脂水平、肝脏指数。采用油红O染色和Van Gieson胶原纤维染色观察颈动脉病理学改变。n 结果:与模型组比较,消瘀降脂胶囊高剂量组小鼠体重降低(n P<0.05),血清TC[(25.92±4.21)mmol/L比(30.39±4.67)mmol/L]、LDL-C[(7.97±2.14)mmol/L比(10.26±1.97)mmol/L]水平降低(n P<0.05),消瘀降脂胶囊低、高剂量组HDL-C[(0.31±0.07)mmol/L、(0.34±0.13)mmol/L比(0.26±0.09)mmol/L]水平升高(n P<0.05)。病理结果显示,阿托伐他汀组及消瘀降脂胶囊高剂量组颈动脉脂质沉积阻塞比例低于模型组(n P<0.05),且尚未形成血管斑块。n 结论:消瘀降脂胶囊可降低ApoEn -/-小鼠血清LDL-C、TC水平,升高HDL-C水平,改善高血脂症状,减轻颈动脉狭窄程度,减缓颈动脉斑块生成。n “,”Objective:To investigate the effects of n Xiaoyu-Jiangzhi capsules on blood lipid, carotid artery atherosclerosis (CAA) and plaque in apolipoprotein E knockout (ApoEn -/-) mice.n Methods:The ApoEn -/- mice were fed with high-fat food to establish carotid atherosclerosis model. The ApoEn -/- mice were randomly by weight divided into model group, Atorvastatin group, low- and high-dose n Xiaoyu-Jiangzhi capsules group. The C57BL/6cnc mice were used as control group and fed with normal diet. The Atorvastatin group was given atorvastatin suspension 1.3 mg/kg, low and high dose groups were given n Xiaoyu-Jiangzhi capsule suspension 325 and 975 mg/kg, and the control group and model group were given equal volume of distilled water. The mice were gavaged with 0.1 ml/10 g body weight, once a day, and the weight of mice was recorded weekly. After 12 weeks of continuous intragastric administration, the blood lipid and liver /body weight index of the mice were measured. Carotid arteries were sliced to conduct oil red O staining and VG staining for the pathological analysis.n Results:After 12 weeks of drug administration, the weight of mice in the high-dose group was significantly lower than the model group. The level of TC (25.92 ± 4.21 mmol/L n vs. 30.39 ± 4.67 mmol/L) and LDL-C (7.97 ± 2.14 mmol/L n vs. 10.26 ± 1.97 mmol/L) in the high-dose group significantly decreased (n P<0.05), the level of HDL-C in the low and high-dose group significantly increased (n P<0.05). The pathological results showed that after 12 weeks of administration, the carotid artery lipid deposition blockage rate in the Atorvastatin group and the high dose group were significantly smaller than the model group(n P<0.05), and no vascular plaque has been formed.n Conclusion:The n Xiaoyu-Jiangzhi capsules could reduce LDL-C, increase HDL-C levels, reduce the constriction of arterial stenosis and slow down the formation process of carotid plaque.n
