目的 研究人食管癌细胞株 EC1和 EC18的核基质形态及蛋白成分。 方法 应用细胞的选择性抽提、DGD包埋 -去包埋剂电镜制样观察核基质形态 ,SDS-聚丙烯酰胺凝胶电泳、双向电泳和免疫印迹等分析核基质蛋白成分。 结果 抽提后 ,两种细胞内均存在精细的核基质 -核纤层 -中间丝网架。低分化的 EC1细胞核基质较高分化的 EC18更细密。核基质蛋白电泳显示 ,两株细胞有数种相同的核基质蛋白成分 ,也有各自的特异性核基质蛋白。在免疫印迹分析中 ,使用抗人 Nu MA单克隆抗体检测到两株食管癌细胞中都存在 Nu MA蛋白。 结论 在食管癌细胞中 ,存在特异的核基质蛋白 ,核基质 -核纤层 -中间丝形成一个连续的体系 ,对维持细胞核的完整性和细胞功能起重要作用。
Objective To study the nuclear matrix and protein composition of human esophageal cancer cell lines EC1 and EC18. Methods The nuclear matrix was observed by selective extraction of cells and embedded in DGD-embedded electron microscope. SDS-polyacrylamide gel electrophoresis, two-dimensional electrophoresis and western blot were used to analyze the nuclear matrix protein components. Results After extraction, fine nuclear matrix-nuclear layer-intermediate silk mesh frame was present in both cells. The poorly differentiated EC1 nuclear matrix has a more differentiated EC18. Nuclear matrix protein electrophoresis showed that the two cells had several identical nuclear matrix protein components and also had their own specific nuclear matrix proteins. In the immunoblot analysis, Nu MA protein was detected in both esophageal cancer cells using an anti-human Nu MA monoclonal antibody. Conclusion There is a specific nuclear matrix protein in the esophageal cancer cells, and the nuclear matrix-nuclear layer-intermediate filament forms a continuous system, which plays an important role in maintaining the integrity and cell function of the nucleus.
