目的 观察膜活化剂吐温 80合并温热作用对人胃癌细胞BGC 82 3的抑制效应及其对Hsp70、bcl 2、bax表达的影响。方法 通过MTT法 ,测定 0 .1%及 0 .15 %吐温 80与 4 2℃温热合并作用对BGC 82 3细胞的抑制作用 ;运用免疫细胞化学染色 ,观察 0 .1%吐温 80与 4 2℃温热合并作用后 (0h、8h、16h、2 4h)BGC 82 3细胞Hsp70、bcl 2、bax表达的改变。结果 吐温 80合并 4 2℃10 0min温热作用对BGC 82 3细胞具有明显的抑制效应 ,温热后保留吐温 80继续作用将进一步增强抑制效应 ,合并抑制效应与吐温 80浓度相关 (P <0 .0 1)。合并作用能明显抑制BGC 82 3细胞热作用后的Hsp70表达 ,但bcl 2、bax的表达均增加 ,且bax的表达强于bcl 2 ,并全部分布于胞浆。结论 吐温 80与 4 2℃温热有明显协同抑制肿瘤细胞生长效应 ,对Hsp70和bcl 2、bax表达及分布的影响提示其抑制机理可能与诱导肿瘤细胞凋亡有关。
Objective To observe the inhibitory effect of membrane activator Tween 80 combined with warming on human gastric cancer cell line BGC 82 3 and its effect on the expression of Hsp70, bcl 2 and bax. Methods MTT assay was used to determine the inhibitory effect of 0.1% Tween 80 and 0.4% Tween 80 on the proliferation of BGC 82 3 cells. Immunocytochemical staining was used to observe the effects of 0.1% Tween 80 and The changes of Hsp70, bcl 2 and bax expression in BGC 82 3 cells were observed after 2 h warm - up (0 h, 8 h, 16 h, 24 h). Results Tween 80 combined with warming at 104 ℃ for 10 min had a significant inhibitory effect on BGC-82 3 cells, and retained Tween-80 after warming further enhanced the inhibitory effect, which was correlated with the concentration of Tween 80 (P <0 .0 1). The combined effect could significantly inhibit the expression of Hsp70 after heat treatment in BGC-82 3 cells, but the expression of bcl 2 and bax increased, and the expression of bax was stronger than that of bcl 2, all of which were distributed in the cytoplasm. Conclusion Tween 80 and warm 4 2 ℃ synergistically inhibit the growth of tumor cells. The effects of Tween 80 and bcl - 2 on the expression and distribution of Hsp70 may be related to the induction of apoptosis.
