以水提醇沉法从荞麦蜂花粉中提取可溶性多糖,采用单因素和正交试验确定多糖提取的最佳工艺条件;通过对邻苯三酚碱性自氧化体系和H2O2/Fe2+体系体外研究荞麦花粉多糖对氧化反应的影响,评价多糖的抗氧化性能。结果表明,荞麦蜂花粉多糖最佳提取工艺为提取温度90℃、提取时间5h、料液比(m/v)1:20、pH为8.0;此工艺条件下多糖平均得率为2.52%。此多糖对O2-.和.OH具有一定的清除作用,当多糖浓度为3.5mg/mL时,对O2-.自由基的清除率为39.41%,是同浓度Vc的0.4倍,对.OH自由基的清除率为65.23%,是同浓度Vc的0.66倍。表明荞麦蜂花粉多糖具有一定的抗氧化性但不及Vc。
Soluble polysaccharides were extracted from the buckwheat pollen by water extraction and alcohol precipitation method. The optimum technological conditions of polysaccharides extraction were determined by single factor and orthogonal experiments. The effects of pyrogallic acid alkaline autoxidation and H2O2 / Fe2 + Pollen polysaccharides on the oxidation reaction, evaluate the antioxidant properties of polysaccharides. The results showed that the optimal extraction process of pollen polysaccharide from buckwheat was extraction temperature 90 ℃, extraction time 5h, ratio of material to liquid (m / v) 1:20 and pH 8.0. The average yield of polysaccharide was 2.52%. This polysaccharide has a certain scavenging effect on O2- and .OH. When the polysaccharide concentration is 3.5mg / mL, the clearance rate of O2- radical is 39.41%, which is 0.4 times of the same concentration Vc, Base clearance rate was 65.23%, 0.66 times the same concentration of Vc. Show that buckwheat bee pollen polysaccharides have some antioxidant but less than Vc.
