目的建立检测实验室中溴乙啶(EB)残量的高效液相色谱电化学法。方法以正己烷为采样载体对PCR实验室中的操作台、仪器载面等区域进行采样,用氧化铝层析柱对样品进行固相萃取,乙腈-冰乙酸溶液作为流动相从Symmetry C8柱上分离和测定EB,并借助于电化学检测器于+780 mV检测。结果标准曲线的线性范围为5.0-500.0 ng/L,平均相对标准偏差为4.8%,检出限为2.9 ng/L,平均回收率为92.2%-104.2%。结论该方法准确、简单、快速、灵敏,适用于EB残量的检测。
OBJECTIVE To establish a high performance liquid chromatography electrochemical detection method for detecting the residual quantity of ethidium bromide (EB) in the laboratory. Methods n-Hexane was used as the sample carrier to sample the area such as operating table and instrument carrier in the PCR laboratory. The samples were subjected to solid-phase extraction with alumina column and the acetonitrile-glacial acetic acid solution as the mobile phase from the Symmetry C8 column EBs were separated and assayed and detected at +780 mV with the aid of an electrochemical detector. Results The calibration curve showed a linear range of 5.0-500.0 ng / L with a mean relative standard deviation of 4.8% and a detection limit of 2.9 ng / L with an average recovery of 92.2% -104. 2%. Conclusion The method is accurate, simple, rapid and sensitive and is suitable for the detection of EB residuals.
