目的体外研究病原体抗原活化的γδT细胞的细胞毒和细胞因子产生的反应性以及对CD4+T细胞增殖的影响,以期进一步澄清γδT细胞的功能。方法用洗淘法从外周血中分离纯化出γδT细胞,在体外探讨植物血凝素和结核分枝杆菌(M.tb)、人类6型疱疹病毒(HHV-6)和阴道毛滴虫(TV)病原体抗原对γδT细胞的细胞毒活性和细胞因子产生的影响,并采用细胞隔离渗透培养法观察γδT细胞分泌的细胞因子对CD4+T细胞增殖的作用。结果M.tb比TV可更强地促进γδT细胞对K562细胞的杀伤活性。M.tb和HHV-6可刺激γδT细胞主要分泌TH1型细胞因子IFN-γ,而TV可刺激γδT细胞优势产生TH2型细胞因子IL-6。经抗原活化的γδT细胞分泌的可溶性因子可促进CD4+T细胞增殖。结论细胞内感染的病原体是激发γδT细胞产生TH1型细胞因子的诱导因子,并对其杀伤活性有所增强,而细胞外感染病原体可激发γδT细胞产生TH2型的细胞因子。
Objective To study the cytotoxicity and cytokine production of γδT cells activated by pathogen antigens and their effects on the proliferation of CD4 + T cells in vitro in order to clarify the function of γδT cells. Methods γδT cells were isolated and purified from peripheral blood by washing-washing method. Phytohemagglutinin and Mycobacterium tuberculosis (M.tb), Human herpesvirus 6 (HHV-6) and Trichomonas vaginalis ) Pathogen antigens on the cytotoxicity and cytokine production of γδT cells. The effects of cytokines secreted by γδT cells on the proliferation of CD4 + T cells were observed by cell isolation and permeabilization. Results M. tb promotes the cytotoxicity of γδT cells to K562 cells more strongly than TV. M. Tb and HHV-6 can stimulate γδ T cells to secrete TH1 type cytokine IFN-γ, while TV can stimulate γδ T cells to produce TH2 type cytokine IL-6. Soluble factors secreted by antigen-activated γδT cells promote CD4 + T cell proliferation. Conclusion The pathogen of intracellular infection is an inducer of TH1-type cytokines induced by γδT cells and its cytotoxicity is enhanced. However, extracellular pathogens can induce γδT cells to produce TH2-type cytokines.
