以贵州玛瑙红樱桃的休眠芽为外植体,通过不同激素质量浓度组合的正交试验,建立了高效的离体培养体系;利用ISSR分子标记,对组培苗的遗传变异进行检测,结果表明:在引物检测范围内,玛瑙红樱桃离体快繁至第8代仍然能保持其遗传稳定性;DNA甲基化的MASP分析表明,组培苗的甲基化敏感多态性随继代次数的增加而逐步升高,第5代组培苗总甲基化率最高,为21.4%,全甲基化率为12.4%,半甲基化率为9.0%.
Using the dormant buds of Guizhou red agar as explants, an efficient in vitro culture system was established by orthogonal test with different combinations of hormone concentration. The ISSR molecular markers were used to detect the genetic variation of tissue culture seedlings. The results showed that : Within the range of primer detection, agate red cherry ex-vivo up to the eighth generation can still maintain its genetic stability; DNA methylation MASP analysis showed that the tissue culture seedling methylation-sensitive polymorphism with the number of generations The fifth generation of tissue culture seedlings had the highest methylation rate of 21.4%, the full methylation rate of 12.4% and the half methylation rate of 9.0%.
