目的:研究花生四烯酸( AA) 环氧化酶产物及5脂氧化酶产物对大鼠加速性肾毒性血清性肾炎( NSN) 中肾小球诱生型一氧化氮合成酶(iNOS) 表达的影响。 方法:制备NSN 大鼠模型后,应用消炎痛抑制AA环氧化酶,应用MK0951 抑制AA 的5脂氧化酶,观察对肾小球iNOS 表达的影响。 结果:在NSN 大鼠,应用消炎痛抑制环氧化酶,可减少肾小球合成的前列腺素(PG) E2 及血栓素( TX) B2 ,使肾小球iNOS 表达增强,尿亚硝酸盐( NO-2 ) 排出增多;应用MK0951 抑制5脂氧化酶,可减少肾小球合成的白三烯(LT) B4 ,也使肾小球iNOS 表达增强,尿NO-2 增多。 结论:NSN 大鼠肾小球合成的环氧化酶及5脂氧化酶产物PGE2 、TX 及LT 有抑制肾小球iNOS 表达的作用。
OBJECTIVE: To study the effects of arachidonic acid (AA) cyclooxygenase and 5-lipoxygenase on glomerular-inducible nitric oxide synthase (iNOS) in rat with accelerated nephrotoxic serum nephritis (NSN) The impact of expression. Methods: NSN rat model was prepared, the application of indomethacin inhibition of cyclooxygenase, the application of MK 0951 inhibition of AA 5 lipoxygenase, glomerular iNOS expression was observed. Results: In NSN rats, the inhibition of cyclooxygenase by indomethacin reduced the synthesis of prostaglandin (PG) E2 and thromboxane B2 in the glomerulus, enhanced the expression of iNOS in glomerulus, increased the expression of urinary nitrite NO-2) increased excretion; application MK 0951 inhibition of 5 lipoxygenase, can reduce glomerular synthesis of leukotriene (LT) B4, but also glomerular iNOS expression increased urinary NO-2 increased. CONCLUSIONS: The cyclooxygenase and 5-lipoxygenase products PGE2, TX and LT synthesized in the glomeruli of NSN rats inhibit the expression of glomerular iNOS.
