利拉鲁肽对大鼠心肌缺血再灌注损伤的保护作用

目的研究利拉鲁肽对大鼠心肌缺血再灌注损伤的保护作用与机制。方法将健康雄性的SD大鼠随机分为3组:假手术组、缺血再灌注组、利拉鲁肽组(70μg·kg-1),每组8只。结扎大鼠冠状动脉前降支30 min、开放120 min,制备心肌缺血再灌注模型。用伊文氏蓝、氯化三苯基四氮唑法测定心肌梗死面积;分别用原位末端标记法及免疫组化法检测心肌细胞凋亡及心肌凋亡蛋白Bcl-2、Bax蛋白的表达。结果利拉鲁肽组与缺血再灌注组相比,心肌梗死面积明显减小(均P<0.05);心肌细胞凋亡数明显减少,Bcl-2蛋白表达明显增高,Bax蛋白表达明显降低(均P<0.05)。结论利拉鲁肽可以通过抑制心肌细胞凋亡来减轻大鼠心肌缺血再灌注损伤。 Objective To study the protective effect and mechanism of liraglutide on myocardial ischemia-reperfusion injury in rats. Methods Healthy male Sprague-Dawley rats were randomly divided into 3 groups: sham operation group, ischemia-reperfusion group and liraglutide group (70μg · kg-1), with 8 rats in each group. Ligation of anterior descending coronary artery 30 min, open 120 min, preparation of myocardial ischemia-reperfusion model. Myocardial infarction area was determined by Evans blue and triphenyltetrazolium chloride method. Apoptosis and expressions of Bcl-2 and Bax protein in cardiomyocytes were detected by in situ end labeling and immunohistochemistry respectively. Results Compared with ischemia / reperfusion group, the area of ​​myocardial infarction in liraglutide group was significantly decreased (all P <0.05), the number of apoptotic cardiomyocytes was significantly decreased, the expression of Bcl-2 protein and Bax protein were significantly decreased All P <0.05). Conclusion Liraglutide attenuates myocardial ischemia-reperfusion injury in rats by inhibiting cardiomyocyte apoptosis.
