目的 探讨健康儿童外周血单核细胞载脂蛋白E基因表达及其与载脂蛋白E基因型之间的关系。方法 采用竞争性逆转录 -聚合酶链反应方法分析 46例正常健康儿童外周血单核细胞的载脂蛋白E基因表达 ,采用改良的聚合酶链反应 -限制性片段长度多态性方法检测载脂蛋白E基因型。结果 载脂蛋白E基因能在健康儿童外周血单核细胞表达 ,健康儿童载脂蛋白E基因表达量为 (0 47± 0 15 )moles/moleGAPDHmRNA ,载脂蛋白E基因表达量没有明显的性别差异 (P >0 0 5 )。健康儿童载脂蛋白E基因型分布不均 ,E 3 /3 (3 2例 ) ,E 2 /3 (8例 )及E4/3(6例 ) ,未见E 2 /2、E 2 /4、E 4/4。载脂蛋白ε 2、ε 3、ε 4等位基因的频率分别是 0 0 9、 0 85、 0 0 6。与ε 3等位基因携带者比较 ,ε 2等位基因携带者载脂蛋白E基因表达量上调 ,ε 4等位基因携带者的载脂蛋白E基因表达量下调〔分别为 (0 47± 0 12 )moles/moleGAPDHmRNA、 (0 42± 0 10 )moles/moleGAPDHmRNA、 (0 2 8± 0 0 9)moles/moleGAPDHmRNA ,P均 <0 0 5〕。结论 载脂蛋白E基因能在健康儿童周围血单核细胞表达 ,载脂蛋白E基因多态性影响健康儿童外周血单核细胞载脂蛋白E基因表达水平。
Objective To investigate the expression of apolipoprotein E gene in peripheral blood mononuclear cells of healthy children and its relationship with apolipoprotein E genotype. Methods The expression of apolipoprotein E in peripheral blood mononuclear cells of 46 healthy children was analyzed by competitive reverse transcription - polymerase chain reaction (PCR). The modified polymerase chain reaction - restriction fragment length polymorphism Protein E genotype. Results Apolipoprotein E gene was expressed in peripheral blood mononuclear cells of healthy children. The expression of apolipoprotein E gene in healthy children was (0 47 ± 0 15) moles / mole GAPDH mRNA, and there was no significant gender difference in apolipoprotein E gene expression (P> 0 0 5). The distribution of apolipoprotein E genotypes in healthy children was unequal, with E 3/3 (3 2 cases), E 2/3 (8 cases) and E 4/3 (6 cases) without E 2/2 and E 2/4 , E 4/4. The frequencies of apolipoprotein ε 2, ε 3 and ε 4 alleles were 0 0 9, 0 85 and 0 0 6, respectively. Compared with carriers of ε 3 allele, ε 2 allele had higher apolipoprotein E gene expression, and ε 4 allele had lower apolipoprotein E gene expression (0 47 ± 0 12) moles / moleGAPDH mRNA, (0 42 ± 0 10) moles / moleGAPDH mRNA, (0 28 ± 0 0 9) moles / moleGAPDH mRNA, all P <0.05). Conclusion Apolipoprotein E gene can be expressed in peripheral blood mononuclear cells in healthy children. Apolipoprotein E gene polymorphism affects the expression of apolipoprotein E in peripheral blood mononuclear cells in healthy children.
