本工作用自旋标记ESR波谱方法研究了HEP.A22鼠腹水肝癌细胞和鼠正常肝细胞膜脂的运动性。采用两种脂肪酸类自旋标记化合物I(12,3)和I(1,14)。在0°—50℃的范围内,每隔5度记录ESR谱。测量了膜脂的旋转相关时间、序参数和摆动半幅度角,并计算了旋转活化能和相转变温度。结果表明:(1) 对癌细胞,其表层膜脂的流动性比正常细胞大,但在膜的深层,则结果相反;(2) 细胞癌变后,其动力学性质亦发生明显改变。在膜表层,癌细胞膜脂的相转变温度为~18℃,正常细胞为~30℃;而在膜的深层,正常细胞的Arrhenius作图表现为一直线,没有观察到转折点;对腹水肝癌细胞,Arrhenius作图不是一条直线,但不排斥多于一个转折点的可能性。
In this work, spin-labeled ESR spectroscopy was used to study the motility of membrane lipids in HEP-A22 murine ascites hepatoma cells and normal rat hepatocytes. Two fatty acid spin labeling compounds I (12,3) and I (1,14) were used. ESR spectra were recorded every 5 degrees in the range of 0 ° -50 ° C. The rotational time, order parameters and half angle of oscillation of membrane lipid were measured. The rotational activation energy and phase transition temperature were calculated. The results showed that: (1) The fluidity of membrane lipids in cancer cells was larger than that in normal cells, but in the deeper layers of membrane, the results were opposite; (2) The dynamic changes of cells were also significantly changed after canceration. In the membrane surface, the phase transition temperature of cancer cell membrane lipid was ~ 18 ℃ and normal cell was ~ 30 ℃. In the deep layer of the membrane, Arrhenius plot of normal cells showed a straight line with no turning point observed. Ascites hepatocellular carcinoma cells, Arrhenius plotting is not a straight line, but does not exclude the possibility of more than one turning point.
