目的:建立祛瘀清热颗粒剂中苦杏仁苷和甘草酸的含量测定方法。方法:采用高效液相色谱法对祛瘀清热颗粒剂中苦杏仁苷和甘草酸进行含量测定。色谱柱:Hypersil BDS C18(5μm,4.6mm×250mm),流速:1.0mL/min,柱温:25℃,进样量:10μL。苦杏仁苷流动相:乙腈-水(17∶83),检测波长为210nm;甘草酸流动相:甲醇-0.2%醋酸铵-冰乙酸(67∶33∶1),检测波长为250nm。结果:苦杏仁苷在2.19~140.00μg/mL范围内线性关系良好(r=0.9999),平均回收率为98.91%,RSD为0.89%(n=9);甘草酸在4.68~149.60μg/mL范围内线性关系良好(r=0.9998),平均回收率为98.35%,RSD为1.01%(n=9)。结论:所建立的HPLC方法专属性强,准确可靠,可用于祛瘀清热颗粒剂中苦杏仁苷和甘草酸的含量测定。
Objective: To establish a method for the determination of amygdalin and glycyrrhizic acid in Quyu Qingre granules. Methods: The contents of amygdalin and glycyrrhizic acid in Quyu Qingre granules were determined by high performance liquid chromatography. Column: Hypersil BDS C18 (5 μm, 4.6 mm × 250 mm), flow rate: 1.0 mL / min, column temperature: 25 ° C, injection volume: 10 μL. Amygdalin mobile phase: acetonitrile-water (17:83), detection wavelength of 210nm; glycyrrhizic acid mobile phase: methanol -0.2% ammonium acetate - glacial acetic acid (67: 33: 1), detection wavelength of 250nm. Results: Amygdalin had a good linearity (r = 0.9999) in the range of 2.19-140.00 μg / mL with an average recovery of 98.91% and a RSD of 0.89% (n = 9). The content of glycyrrhizin in the range of 4.68-149.60 μg / mL The linear relationship was good (r = 0.9998). The average recovery was 98.35% with RSD of 1.01% (n = 9). Conclusion: The established HPLC method is specific, accurate and reliable, and can be used for the determination of amygdalin and glycyrrhizin in Quyu Qingre granules.
