采用盐酸胍/氯化铯超离法及Oligo(dT)柱层析法从日本血吸虫成虫中成功地分离出完整的mRNA。血吸虫mRNA在兔网状细胞溶解产物中能翻译蛋白质,使体外合成的蛋白质中(35)~S-甲硫氨酸的掺入增加。对流免疫电泳放射自显影的结果表明,血吸虫mRNA的翻译产物与感染兔血清产生沉淀,提示抗血吸虫血清能特异地识别上述蛋白质的决定簇。血吸虫mRNA的抽提成功为以后克隆日本血吸虫抗原的基因打下了基础。
The complete mRNA was successfully isolated from Schistosoma japonicum adults using guanidine hydrochloride / cesium chloride super-isolation and Oligo (dT) column chromatography. Schistosoma japonicum mRNA translates proteins in rabbit reticulocyte lysates, resulting in increased incorporation of (35) S-methionine into proteins synthesized in vitro. The results of autoradiography of convective immunoelectrophoresis showed that the transgene of Schistosoma japonicum mRNA and the sera of infected rabbits were precipitated, suggesting that the serum of schistosoma japonicum can specifically recognize the determinants of these proteins. Schistosoma japonicum mRNA extraction success for the subsequent cloning of Schistosoma japonicum antigen gene laid the foundation.
