目的探讨葡萄糖转运蛋白1(GLUT-1)和己糖激酶-Ⅱ(HK-Ⅱ)蛋白在乳腺癌组织中的表达及其预测患者预后的价值。方法选取2016年1月至2016年7月行手术治疗的乳腺癌患者32例为研究对象,术后随访至少半年。术后均采用免疫组织化学法行乳腺癌组织和癌旁组织GLUT-1和HK-Ⅱ蛋白表达的检测,比较不同预后患者的乳腺癌组织GLUT-1和HK-Ⅱ蛋白表达情况,分析其表达与预后的关系,进一步评价其预测患者预后的价值。结果32例患者随访期间的复发转移率为31.25%。乳腺癌组织GLUT-1蛋白阳性表达率(65.63%)及HK-Ⅱ蛋白阳性表达率(68.75%)明显高于其癌旁组织(9.38%,18.75%;P均<0.01);复发转移患者乳腺癌组织GLUT-1蛋白阳性表达率(100.00%)和HK-Ⅱ蛋白阳性表达率(100.00%)均高于无复发转移患者(50.00%,54.55%;P均<0.05)。Logistic回归分析结果显示,乳腺癌组织GLUT-1和HK-Ⅱ蛋白表达与其复发转移均密切相关(P均<0.01)。诊断性试验四格表分析结果显示,乳腺癌组织GLUT-1蛋白表达预测其复发转移的敏感度、特异度、准确性分别为47.62%、100.00%、65.63%,HK-Ⅱ蛋白表达分别为45.45%、100.00%、62.50%,两者联合预测分别为86.36%、100.00%、90.63%。结论乳腺癌组织GLUT-1和HK-Ⅱ蛋白表达升高且两者联合预测患者预后的价值良好,可能作为乳腺癌预后评估指标和临床治疗靶点。
Objective To investigate the expression of glucose transporter 1 (GLUT-1) and hexokinase-Ⅱ (hk-Ⅱ) in breast cancer and their prognostic value. Methods Thirty-two patients with breast cancer who underwent surgical treatment from January 2016 to July 2016 were selected as the study subjects and were followed up for at least six months. The expression of GLUT-1 and HK-Ⅱ protein in breast cancer tissues and paracancer tissues were detected by immunohistochemistry after operation, and the expressions of GLUT-1 and HK-Ⅱ protein in breast cancer tissues were compared between different prognosis groups And the prognosis of the relationship between further evaluation of the prognosis of patients with the value of prognosis. Results The recurrence and metastasis rate of 32 patients during follow-up was 31.25%. The positive rate of GLUT-1 protein (65.63%) and the positive rate of HK-Ⅱ protein (68.75%) in breast cancer tissues were significantly higher than those in paracancerous tissues (9.38%, 18.75%; P <0.01) The positive rate of GLUT-1 protein (100.00%) and the positive rate of HK-Ⅱ protein in cancer tissues (50.00%, 54.55%, P <0.05) were higher than those without recurrence and metastasis (100.00%). Logistic regression analysis showed that the expression of GLUT-1 and HK-Ⅱ in breast cancer was closely related to the recurrence and metastasis (all P <0.01). Diagnostic test of four grid table analysis showed that the sensitivity, specificity and accuracy of GLUT-1 protein expression in breast cancer were 47.62%, 100.00% and 65.63%, respectively, and the expression of HK-Ⅱ protein was 45.45 %, 100.00% and 62.50%, respectively. The joint prediction is 86.36%, 100.00% and 90.63% respectively. Conclusions The expression of GLUT-1 and HK-Ⅱ protein in breast cancer is increased, and the combination of the two is of good value in predicting prognosis of patients with breast cancer, which may serve as a prognostic indicator and clinical target for breast cancer.
