目的研究石英诱发永生化的人支气管上皮细胞系(16HBE)的恶性转化作用。方法用新研磨的浓度为300 mg/L的石英染毒16HBE细胞共10次,软琼脂集落形成实验和裸鼠成瘤实验鉴定转化细胞的恶性程度。结果细胞培养至35代,发现石英可以诱导16HBE发生恶性转化。转化细胞排列紊乱,重叠生长,在软琼脂中的集落形成率显著高于对照组(P<0.01);转化细胞在裸鼠体内成瘤。结论石英具有较强的诱导16HBE恶性转化能力。
Objective To investigate the malignant transformation of quartz immortalized human bronchial epithelial cell line (16HBE). Methods 16HBE cells were treated with freshly ground quartz at a concentration of 300 mg / L for 10 times. Soft agar colony formation assay and nude mouse tumorigenicity assay were used to identify the malignant degree of transformed cells. Results The cells were cultured to 35 passages and quartz was found to induce malignant transformation of 16HBE. The transformed cells were disordered and overlapped. The colony formation rate in soft agar was significantly higher than that in control group (P <0.01). The transformed cells formed tumors in nude mice. Conclusion Quartz has a strong ability to induce malignant transformation of 16HBE.
