目的建立高效液相色谱法(high performance liquid chromatography,HPLC)测定左乙拉西坦血药浓度的方法。方法血清样品经液-液萃取处理。色谱柱为Diamonsil C18(250×4.6mm,5μm),流动相为乙腈-0.4%磷酸溶液(20:80,V/V),检测波长210nm,柱温35℃,流速1.0mL/min。结果左乙拉西坦的保留时间为4.3min,内标保留时间为8.4min,无干扰;线性关系良好,线性范围1~80mg/L。结论该方法简便、准确,可用于临床测定左乙拉西坦的血药浓度,指导临床合理用药。
Objective To establish a method for the determination of levetiracetam plasma concentration by high performance liquid chromatography (HPLC). Methods Serum samples were subjected to liquid-liquid extraction. The chromatographic column was Diamonsil C18 (250 × 4.6 mm, 5 μm). The mobile phase was acetonitrile-0.4% phosphoric acid solution (20:80, V / V). The detection wavelength was 210 nm. The column temperature was 35 ℃ and the flow rate was 1.0 mL / min. Results The retention time of levetiracetam was 4.3 min, the retention time of internal standard was 8.4 min, without interference; the linearity was good with a linear range of 1 ~ 80 mg / L. Conclusion The method is simple and accurate, which can be used for clinical determination of plasma concentration of levetiracetam and guide clinical rational drug use.
