目的研究非毒性分化诱导剂苯了酸钠(phenylbutyrate,NaPB)在体外对人脑胶质细胞瘤BT325细胞的生长、分化的影响。方法应用形态学观察法、MTT分析法、流式细胞仪对在体外经不同浓度的NaPB处理过的BT325细胞进行检测。结果1.电子显微镜观察,在NaPB处理过的BT325细胞中,线粒体、内质网、弹力丝丰富,而在NaPB未处理的细胞中,有大量游离核糖体,上述细胞结构却未能观察到;2.NaPB在2mM时就能明显抑制BT325细胞的生长.并且有时间依赖关系及剂量依赖关系;3.应用流式细胞仪测定发现,经NaPB处理的细胞其S期在11.99%或13.17%.而未经处理的细胞其S期在20.17%。结论NaPB在体外不仅能够抑制人脑胶质细胞瘤的生长,而且能诱导细胞的分化,为NaPB分化治疗胶质瘤提供了实验依据。
Objective To investigate the effect of nontoxic differentiation inducer phenylbutyrate (NaPB) on the growth and differentiation of human glioblastoma BT325 cells in vitro. Methods Morphological observation, MTT assay and flow cytometry were used to detect BT325 cells treated with different concentrations of NaPB in vitro. Results 1. Electron microscope observation showed that mitochondria, endoplasmic reticulum, and elastic filaments were abundant in NaPB-treated BT325 cells, but there were a large number of free ribosomes in NaPB-untreated cells, but the above cell structures were not observed. NaPB can significantly inhibit the growth of BT325 cells at 2 mM. And there is a time-dependent and dose-dependent relationship; 3. Flow cytometric analysis revealed that the S phase of NaPB-treated cells was 11.99% or 13.17%. The untreated cells had an S phase of 20.17%. Conclusion NaPB not only inhibits the growth of human glioma in vitro, but also induces the differentiation of cells, providing an experimental basis for NaPB differentiation and treatment of glioma.
