目的研究食管癌患者外周血中TACSTD1和CK19的表达,探讨其作为食管癌微转移标志物的可行性和其对食管癌患者预后的影响。方法用实时荧光定量RT-PCR联合检测30例食管癌患者手术切除的肿瘤标本和食管残端TACSTD1和CK19的表达并比较二者的区别,检测30例健康献血员外周血标本TACSTD1和CK19的表达得出正常人群的表达范围,以此为标准检测30例食管癌患者术前外周血标本是否存在TACSTD1和/或CK19的高表达。结果 TAC-STD1和CK19在食管癌组织中的表达量均明显高于食管残端组织(P<0.01)。22例患者的外周血标本检测出TACSTD1和/或CK19的高表达。CK19和TACSTD1在外周血中的表达量与患者的肿瘤分化程度高低(P<0.05)和T分期(P<0.05)、N(P<0.01)分期有密切联系。结论 CK19和TACSTD1在食管癌组织均有特异性的高表达,在食管癌的发生、发展中可能起着重要的作用。CK19和TACSTD1是有价值的食管癌微转移标志物。
Objective To study the expression of TACSTD1 and CK19 in the peripheral blood of patients with esophageal cancer and to investigate their feasibility as micrometastasis markers of esophageal cancer and their effects on the prognosis of patients with esophageal cancer. Methods The expression of TACSTD1 and CK19 in 30 resected esophageal cancer specimens and TACSTD1 and CK19 in esophageal cancer patients were detected by real-time fluorescence quantitative RT-PCR. The expression of TACSTD1 and CK19 in 30 healthy blood donors The normal expression of the population was obtained as a standard test of 30 cases of esophageal cancer patients with preoperative peripheral blood samples for the presence of TACSTD1 and / or CK19 high expression. Results The expression of TAC-STD1 and CK19 in esophageal cancer tissue was significantly higher than that in esophageal stump tissue (P <0.01). Peripheral blood samples of 22 patients detected high expression of TACSTD1 and / or CK19. The expression of CK19 and TACSTD1 in peripheral blood was closely related to the degree of tumor differentiation (P <0.05) and T stage (P <0.05), and N (P <0.01) stages in patients. Conclusion Both CK19 and TACSTD1 are highly expressed in esophageal cancer tissues and may play an important role in the development of esophageal cancer. CK19 and TACSTD1 are valuable micrometastatic markers of esophageal cancer.
