目的 :研究气味沙雷氏菌粗核糖体制剂 (RP)中的抗肿瘤活性成分。方法 :以RP的多克隆抗血清做Western blot ,寻找制剂中特异性的免疫原 ;以DEAE 32离子交换层析法结合SephadexG 2 0 0层析法纯化之 ,体外细胞毒试验快速鉴定其抑瘤活性。结果 :RP及盐洗核糖体制剂 (SR)有两种特异性的免疫原 ,分子量分别为 16 .5kDa和 12kDa ,而层析核糖体制剂 (CR)中却没有 ;纯化的 16 .5kDa蛋白质印渍反应与RP中 16 .5kDa处的印渍反应一致 ;体外细胞毒性试验发现它对P388细胞的生长有抑制作用 ,IC50 可达 0 .0 0 1g/L ,但它不影响WISH细胞的生长。结论 :分子量为 16 .5kDa蛋白质是气味沙雷氏菌核糖体制剂体外抑瘤作用的有效成分。
Objective: To study the antitumor activity of Serratia marcescens crude ribosome preparations (RP). Methods: The polyclonal antiserum against RP was used to do Western blot to find the specific immunogen in the preparation. The recombinant protein was purified by DEAE 32 ion exchange chromatography and Sephadex G 20 0 chromatography. active. RESULTS: There were two specific immunogens for RP and salt-washed ribosome preparations (SR) with molecular weights of 16.5 kDa and 12 kDa, respectively, but not in the chromatographic ribosome preparation (CR); the purified 16.5 kDa protein The stain reaction was consistent with the 16.5-kDa imprinting reaction at RP. In vitro cytotoxicity assay showed that it inhibited the growth of P388 cells with an IC50 up to 0.010 g / L, but it did not affect the growth of WISH cells. Conclusion: The molecular weight of 16.5 kDa protein is an effective component of anti-tumor effect of Serratia marcescens ribosome preparation in vitro.
