目的比较含天然牛黄、体培牛黄或人工牛黄的岗藿抗感汤对感染H1N1流感病毒小鼠的保护作用。方法 96只小鼠随机分为正常组、模型组、利巴韦林组、人工牛黄组、天然牛黄组、体培牛黄组,每组16只。除正常组外其余各组小鼠采用H1N1型流感病毒液滴鼻感染小鼠。感染4h后利巴韦林组给予利巴韦林片溶液(浓度3.75 mg/ml)0.2 ml/10 g,各牛黄组分别给予含不同来源牛黄的岗藿抗感汤(浓度2.31 g/ml)0.2 ml/10 g,正常组和模型组灌胃等体积生理盐水,连续给药15天。比较各组小鼠平均存活时间、死亡保护率和生命延长率,肺指数及肺湿干重比,肺组织病毒相对表达量。结果模型组平均存活时间较正常组明显缩短,肺指数、肺湿干重比及肺组织病毒相对表达量升高(P<0.01);与模型组比较,利巴韦林组、人工牛黄组、体培牛黄组和天然牛黄组大鼠存活时间显著延长,肺指数、肺湿干重比及肺组织病毒相对表达量降低(P<0.05或P<0.01)。各牛黄组之间各指标比较差异无统计学意义(P>0.05)。结论含天然牛黄、体培牛黄或人工牛黄的岗藿抗感汤均可延长H1N1病毒感染小鼠存活时间,降低肺组织中病毒载量,牛黄来源的差异不影响岗藿抗感汤对流感病毒感染小鼠的保护作用。
OBJECTIVE To compare the protective effect of Genghuo Kangni Decoction containing natural bezoar, cultured bezoar or artificial bezoar against mice infected with H1N1 virus. Methods Ninety-six mice were randomly divided into normal group, model group, ribavirin group, artificial bezoar group, natural bezoar group and body-cultured group, 16 rats in each group. Except the normal group, mice in other groups were infected with H1N1 influenza virus nasal drops. The ribavirin group was given ribavirin solution (concentration 3.75 mg / ml) 0.2 ml / 10 g 4 h after infection, and each group received different doses of Niuhuang Agmatine (2.31 g / ml) 0.2 ml / 10 g, normal group and model group were given the same volume of normal saline, continuous administration for 15 days. The mean survival time, the rate of death protection, the rate of life prolongation, the lung index and the ratio of wet to dry weight of lung and the relative expression of lung virus in each group were compared. Results Compared with the model group, the average survival time of the model group was significantly shorter than that of the normal group (P <0.01). The lung index, the ratio of lung wet weight to dry weight and the relative expression of pulmonary virus were increased (P <0.01) Survival time was significantly prolonged in body weight Niuhuang group and natural Niuhuang group, and lung index, lung wet / dry weight ratio and lung tissue relative expression were decreased (P <0.05 or P <0.01). There was no significant difference between each group (P> 0.05). CONCLUSION: Nigella, body-cultured or artificial bezoar can both prolong the survival time of mice infected with H1N1 virus and reduce the viral load in lung tissue. The difference of the source of bezoar did not affect the effect of anti-flu virus Infection in mice protective effect.
