目的:建立鸡胚绒毛尿囊膜(CAM)实验模型,观察鬼臼毒素及其衍生物对CAM新生血管生成的影响。方法:鸡胚孵育7d后,将无水乙醇溶解的鬼臼毒素及其衍生物的明胶海绵载体植入CAM上,并孵化72h。剪下CAM,解剖镜下拍照,计数血管,分析其对于血管生成的影响。结果:去氧鬼臼毒素、苦鬼臼毒酮、苦鬼臼毒素和鬼臼毒素具有明显抑制CAM血管生成作用,鬼臼毒酮和异苦鬼臼毒素抑制作用不明显。结论:鬼臼毒素及其衍生物抗肿瘤活性可能与其抑制新生血管的作用有关。
Objective: To establish an experimental model of chick chorioallantoic membrane (CAM) to observe the effect of podophyllotoxin and its derivatives on CAM neovascularization. Methods: After incubating for 7 days, gelatin sponge carrier of podophyllotoxin and its derivatives dissolved in absolute ethanol was implanted into CAM and incubated for 72 hours. Cut the CAM, dissect the microscope and count the blood vessels to analyze their effects on angiogenesis. RESULTS: Deoxypodophyllotoxin, picropodophyllotoxin, picropodophyllotoxin and podophyllotoxin had a significant inhibitory effect on CAM angiogenesis. The inhibitory effect of podophyllotoxin and isophytoflavin was not obvious. Conclusion: The anti-tumor activity of podophyllotoxin and its derivatives may be related to the inhibition of neovascularization.
