目的利用酶联免疫吸附(ELISA)技术检测不同人群血浆中C1qC的蛋白表达水平以及结核病人胸水和血浆C1qC的蛋白表达水平。通过比较C1qC在不同人群和不同标本中的表达情况,评价其在结核诊断中的应用价值。方法利用ELISA技术分别对20例健康对照(HC)、20例结核潜伏感染者(LTBI)和27例肺结核病人(TB)血浆C1qC的表达水平;随访其中10例抗结核治疗的结核病人,检测其血浆中C1qC的表达,观察在抗结核治疗过程中(抗结核治疗前、抗治疗3个月和6个月)C1q C的表达水平;检测16例结核性胸膜炎病人配对的血浆和胸水上清中C1qC的表达水平,比较不同样本中C1qC的表达水平。结果 C1qC在结核健康对照、潜伏感染者、结核病人血浆中的表达水平分别为603.8±134.3、507.0±141.2、756.6±261.2(pg/mL)。肺结核病人C1qC的表达水平显著高于健康对照(P<0.05)和潜伏感染者(P<0.001);在10例结核病人中,C1qC在抗结核治疗前的表达水平为769.6±398.8(pg/mL),抗结核治疗3个月和6个月后C1q C表达分别为624.4±208.5和444.8±163.8(pg/mL),在抗结核治疗6个月后C1qC的表达水平显著地低于抗结核治疗前的水平(P<0.05);C1qC在胸水上清中的表达水平显著高于血浆中的表达水平(P<0.05)。结论 C1qC在结核患者的血浆中高表达,在抗结核治疗中逐渐下降,并且在结核感染部位表达显著增高。这些结果提示利用ELISA检测血浆和胸水上清中C1q C的表达可用于肺结核的辅助诊断。
Objective To detect the expression of C1qC in plasma and the expression of C1qC protein in pleural effusion and plasma of patients with enzyme-linked immunosorbent assay (ELISA). By comparing the expression of C1qC in different populations and different specimens, evaluate its value in the diagnosis of tuberculosis. Methods The plasma levels of C1qC in 20 healthy controls (HC), 20 LTBI patients and 27 pulmonary tuberculosis patients (TB) were detected by ELISA. Ten TB patients with tuberculosis were followed up and tested The expression of C1qC in plasma was observed. The expression level of C1q C in anti-TB treatment (anti-TB treatment, anti-TB treatment 3 months and 6 months) was observed. In the paired plasma and pleural fluid supernatant from 16 patients with tuberculous pleurisy C1qC expression levels in different samples compared C1qC expression levels. Results The expression levels of C1qC in tuberculosis healthy controls, latent infection and tuberculosis patients were 603.8 ± 134.3, 507.0 ± 141.2 and 756.6 ± 261.2 (pg / mL), respectively. The expression level of C1qC in tuberculosis patients was significantly higher than that in healthy controls (P <0.05) and latent infection (P <0.001). In 10 TB patients, the expression level of C1qC before anti-tuberculosis treatment was 769.6 ± 398.8 (pg / mL ), C1q C expression was 624.4 ± 208.5 and 444.8 ± 163.8 (pg / mL) at 3 months and 6 months after anti-TB treatment respectively. The expression level of C1qC was significantly lower than anti-TB treatment 6 months after anti-TB treatment (P <0.05). The expression level of C1qC in pleural fluid supernatant was significantly higher than that in plasma (P <0.05). Conclusions C1qC is highly expressed in the plasma of patients with tuberculosis, gradually declines in the anti-TB treatment, and is significantly increased in the tuberculosis infection site. These results suggest that the use of ELISA to detect C1q C expression in plasma and pleural fluid supernatant can be used for the diagnosis of pulmonary tuberculosis.
