通过建立体外肿瘤坏死因子(TNF-α)诱导人脐静脉血管内皮细胞(HUVEC)炎症损伤反应模型,研究蓝莓叶中槲皮素及其衍生物金丝桃苷对人脐静脉内皮细胞中血管紧张素Ⅰ转化酶(ACE)以及炎症因子(单核细胞趋化蛋白-1、细胞间粘附分子-1和血管细胞间粘附分子-1)含量的影响,采用酶联免疫吸附剂测定方法(ELISA)检测细胞上清液中可溶性ACE和炎症因子蛋白含量的变化,探讨二者在血管内皮细胞中的舒血管和抗炎作用。研究结果表明,肿瘤坏死因子TNF-α可引起HUVEC内ACE和炎症因子含量显著增加,槲皮素和金丝桃苷预处理内皮细胞能显著抑制ACE酶活性和炎症因子含量的增加(p<0.05),具有一定的抗炎和舒血管作用。
To investigate the inflammatory response of human umbilical vein endothelial cells (HUVECs) induced by tumor necrosis factor (TNF-α) in vitro and to investigate the effect of hyperoside in the blueberry leaves and hyperoside on its vascular hypertrophy in human umbilical vein endothelial cells (ACE) and inflammatory cytokines (monocyte chemoattractant protein-1, intercellular adhesion molecule-1 and intercellular adhesion molecule-1) were detected by enzyme-linked immunosorbent assay ELISA) to detect the content of soluble ACE and inflammatory cytokines in the supernatant of the cells, and to explore their vasodilatation and anti-inflammatory effects in vascular endothelial cells. The results showed that tumor necrosis factor TNF-α induced a significant increase of ACE and inflammatory cytokines in HUVECs. Pretreatment with quercetin and hyperin could significantly inhibit the increase of ACE activity and inflammatory cytokines (p <0.05 ), Has a certain anti-inflammatory and vasodilative effect.
