基质生物学的研究使传统的细胞培养技术有重大进展,这种改革包括无血清培养,为每种细胞补充一定的激素和利用组织特异的细胞外基质作为培养底物。这种培养方法能促进细胞附着、克隆生长和细胞分化,尤其适合于上皮细胞的生长。用于细胞培养的基质有胶元、细胞外基质(ECM)粗提物、生物基质、重建基底膜基质、羊膜粗提取物等,分别介绍了大概的提制方法和应用情况。
Research in stroma biology has led to significant advances in traditional cell culture technologies, including serum-free culture, the recruitment of certain hormones to each cell, and the use of tissue-specific extracellular matrix as a substrate for culture. This culture method can promote cell attachment, clonal growth and cell differentiation, especially for epithelial cell growth. The substrates used for cell culture are collagen, extracellular matrix (ECM) crude extract, biological substrate, reconstructed basement membrane matrix and crude amniotic extract. The general methods and application of extraction are introduced respectively.
