[方法]越南伯克霍尔德氏菌B418是优良的生物控制剂,在前期研究中其主要应用剂型为可湿性粉剂,但其保存期较短,而微胶囊的应用可解决该问题。利用双流体气流式雾化器制备越南伯克霍尔德氏菌B418的微胶囊,采用单因子实验研究微胶囊制备的合适条件,利用经典加速实验确定微胶囊的保存期。[结果]微胶囊最佳制备条件包括:壁材海藻酸钠20~30 g/L,固化剂氯化钙40~50 g/L,芯材B418菌液1.0×1012 cfu/L,搅拌速度150~200 r/min,保护剂脱脂奶粉500 g/L,乳化剂吐温80 5 g/L,微胶囊释放液为pH值6.8的磷酸缓冲液。在25℃下保存677 d,微胶囊中菌株B418的存活率仍在90%以上。[结论]微胶囊是适合菌株B418的剂型,该剂型的保存期接近2年,完全满足大生产的需求。
[Method] Burkholderia B418 from Vietnam was an excellent biocontrol agent. In the previous studies, its main application form was WP, but its shelf life was short. However, the application of microcapsules could solve this problem. The microcapsules of Burkholderia B418 from Vietnam were prepared by two-fluid flow nebulizer. The suitable conditions for the preparation of microcapsules were studied by single factor experiment. The storage period of microcapsules was determined by classical accelerating experiment. [Result] The optimum preparation conditions of microcapsules included: 20 ~ 30 g / L sodium alginate for wall material, 40 ~ 50 g / L calcium chloride as curing agent, 1.0 × 1012 cfu / L as core material B418, 150 ~ 200 r / min, protective agent skim milk powder 500 g / L, emulsifier Tween 80 5 g / L, microcapsule release solution pH 6.8 phosphate buffer. When stored at 25 ℃ for 677 days, the viability of strain B418 in microcapsules was still over 90%. [Conclusion] The microcapsule is a suitable dosage form for the strain B418. The storage period of this dosage form is close to 2 years, which fully meets the needs of large-scale production.
