胡桃醌对卵巢癌SKOV3细胞周期影响

目的观察胡桃醌对人卵巢癌SKOV3细胞增殖及细胞周期的影响。方法采用MTT法检测胡桃醌对卵巢癌SKOV3细胞生长抑制作用;采用流式细胞仪检测胡桃醌对卵巢癌SKOV3细胞周期的影响;采用Western Blot检测卵巢癌SKOV3细胞Cyclin D1和Cyclin E的表达。结果与对照组比较,不同剂量胡桃醌对卵巢癌SKOV3细胞具有抑制作用且呈剂量依赖性,IC50为30.13μmol/L,将细胞周期阻滞在G0/G1期。与对照组比较,胡桃醌处理组细胞周期蛋白Cyclin D1表达降低(P<0.05),而Cyclin E蛋白表达变化不显著。结论胡桃醌通过抑制Cyclin D1的表达抑制细胞周期G0/G1期阻滞,抑制细胞生长。 Objective To observe the effects of juglone on the proliferation and cell cycle of human ovarian cancer SKOV3 cells. Methods The inhibitory effect of juglone on SKOV3 ovarian cancer cells was detected by MTT assay. The effect of juglone on the cell cycle of ovarian cancer SKOV3 cells was detected by flow cytometry. The expression of Cyclin D1 and Cyclin E in ovarian cancer SKOV3 cells was detected by Western Blot. Results Compared with the control group, different doses of juglone could inhibit ovarian cancer SKOV3 cells in a dose-dependent manner with the IC50 of 30.13 μmol / L and the cell cycle arrest in G0 / G1 phase. Compared with the control group, the expression of Cyclin D1 in the Juglone group decreased (P <0.05), while the expression of Cyclin E protein did not change significantly. Conclusion Juglone can inhibit the cell cycle arrest and inhibit the cell growth by inhibiting the expression of Cyclin D1.
