目的 探讨针对人RON基因跨膜区段(RONm)反义核酸对肺癌细胞系 A549 活性和细胞增殖的抑制作用,研究以RON基因为靶的肺癌基因治疗的可能性。方法 将人 RONm cDNA反向插入到真核表达载体 pcDNA3.1中,转染肺癌细胞A549,利用ELISA检测细胞模型RON基因的表达水平,同时利用MTT方法监测细胞生物学活性的变化。结果 转染后的肺癌细胞 RON基因表达量和细胞活性明显降低,并出现明显的细胞凋亡。结论 RONm反义核酸明显抑制 RON基因的表达,同时对肺癌细胞的生长有抑制作用,促进细胞凋亡。
Objective To investigate the inhibitory effect of RONm antisense oligonucleotide against A549 cells and cell proliferation in lung cancer cell line A549 and investigate the possibility of gene therapy targeting lung cancer with RON gene. Methods The human RONm cDNA was inserted into the eukaryotic expression vector pcDNA3.1 and transfected into A549 cells. The expression of RON gene was detected by ELISA, and the changes of cell biological activity were detected by MTT assay. Results After transfection, the expression level of RON gene and the cell viability of lung cancer cells were significantly decreased, and obvious apoptosis was observed. Conclusion RONm antisense RNA can obviously inhibit the expression of RON gene and inhibit the growth of lung cancer cells and promote the apoptosis of lung cancer cells.
