目的:研究养血活血方对便秘老年小鼠肠神经递质的影响,阐明养血活血方治疗老年性便秘的作用机理。方法:选取健康老年小鼠,采用吗啡复制便秘小鼠模型,造模成功后,将60只健康老年ICR小鼠随机分为6组:空白组、模型组、阳性对照组(麻仁软胶囊)、养血活血方高、中、低剂量组。各组分别给药,连续灌服3周后处死,取结肠组织。采用免疫组化方法测定肠神经递质SP、VIP、5-HT的表达。结果:与空白组、阳性对照组及养血活血方各剂量组相比,模型组肠壁组织内SP、VIP、5-HT阳性表达明显减少,差异均有显著性统计学意义(P<0.01);各给药组组间比较可见:养血活血方高、中剂量组的神经递质SP、VIP、5-HT含量高于养血活血方低剂量、阳性对照组,差异有统计学意义(P<0.05或P<0.01)。结论:养血活血方可能通过增加肠神经递质SP、VIP、5-HT阳性表达来起到通便的作用,并且养血活血方的通便作用呈一定的量效依赖关系。
Objective: To study the effect of Yangxue Huoxue Recipe on enteron neurotransmitter in senile mice with constipation and clarify the mechanism of Yangxue Huoxue Fang in treating senile constipation. Methods: The healthy aged mice were selected and the mice model of constipation was reproduced by morphine. After successful establishment of model, 60 healthy aged ICR mice were randomly divided into 6 groups: blank group, model group, positive control group (Maren soft capsule) Nourishing blood and activating blood high, medium and low dose group. Each group were administered separately, 3 weeks after continuous oral administration of sacrifice, take the colon tissue. Immunohistochemistry was used to determine the expression of intestinal neurotransmitters SP, VIP and 5-HT. Results: The positive expression of SP, VIP and 5-HT in model group were significantly decreased compared with the blank control group, positive control group and nourishing blood and activating blood prescription. The difference was statistically significant (P <0.01) ); The comparison of each administration group shows that the contents of neurotransmitter SP, VIP and 5-HT of high and middle dose of Yangxue Huoxue Fang are higher than those of low dose and positive control group of Yangxue Huoxue Fang, the difference is statistically significant (P <0.05 or P <0.01). Conclusion: Yangxuehuoxue Prescription can play a role of catharsis by increasing the positive expression of enterotransmitter SP, VIP and 5-HT, and the effect of nourishing blood and promoting blood circulation on purgation is dose-effect dependent.
