目的探讨给予哈伯因(HupA)对急性水胺硫磷中毒有无预防性治疗作用及对乙酰胆碱酯酶(AChE)活力的影响。方法40只小鼠均经口染毒(水胺硫磷22.05 mg/kg)后,随机分为未治疗组、阿托品治疗组、HupA治疗组和HupA与阿托品联合治疗组,每组10只。比较各组中毒症状、死亡率,并测定HupA治疗组和未治疗组全血、红细胞和脑AChE活力。结果HupA干预处理,使水胺硫磷染毒小鼠的中毒症状明显改善,存活率增加,联合治疗组效果最好。水胺硫磷染毒后,HupA治疗组和未治疗组AChE活力值均降低,在染毒后的0.5、1、2h几个时点,HupA治疗组的全血AChE活力(1.096±0.111)、(1.262±0.146)、(1.181±0.353)U/ml,红细胞(0.798±0.063)、(1.000±0.176)、(0.837±0.331)U/ml及脑组织(13.739±2.970)、(18.507±3.466)、(10.764±2.212)U/g均高于未治疗组,差异有统计学意义(P<0.05或P<0.01)。结论HupA对水胺硫磷中毒小鼠有预防性治疗作用,HupA对全血和脑组织中AChE有一定的保护作用,这可能是HupA预防性治疗水胺硫磷中毒作用机制之一。
Objective To investigate the effect of HupA on the preventive treatment of acute acetoside poisoning and the effect on acetylcholinesterase (AChE) activity. Methods Forty mice were randomly divided into untreated group, atropine group, HupA group and HupA combined with atropine group (n = 10) . Toxicity and mortality of each group were compared, and the activity of AChE in whole blood, erythrocytes and brain of HupA treated group and untreated group was determined. Results Intervention with HupA resulted in a significant improvement of the symptoms of poisoning in metoclopramide-treated mice and an increase in survival rate. The combined treatment group had the best effect. AChE activity of HupA treated group and untreated group decreased after exposure to isathionine. At 0.5, 1, 2 hours after HupA treatment, whole blood AChE activity (1.096 ± 0.111), (1.262 ± 0.146), (1.181 ± 0.353) U / ml, erythrocyte (0.798 ± 0.063), (1.000 ± 0.176) (0.837 ± 0.331) U / ml and brain tissue (13.739 ± 2.970), (18.507 ± 3.466) and (10.764 ± 2.212) U / g In untreated group, the difference was statistically significant (P <0.05 or P <0.01). Conclusions HupA has a preventive effect on the mice in the treatment of acetanil poisoning. HupA has a protective effect on AChE in whole blood and brain tissue, which may be one of the mechanisms of HupA prophylaxis in the treatment of acetanilamide poisoning.
