海地瓜中神经酰胺类化合物的分离鉴定及其活性研究

目的:分离、鉴定海地瓜中的一脂类化合物,测定其体外抗肿瘤活性。方法:对海地瓜干粉用氯仿-甲醇-水体系(体积比4∶4∶1)萃取,经连续正反相柱层析分离得到1种脂类化合物。采用IR、ESI-MS、NMR、GC-MS等分析手段对该化合物结构进行表征,并用MTT法检测其对S-180细胞增殖的影响。结果表明:该活性脂质为神经酰胺类化合物系列物,主要由2-氨基-1,3-二羟基-15-甲基-4-十七烯长链碱和5种饱和及单不饱和的脂肪酸构成。MTT试验表明其对S-180小鼠肉瘤细胞的增殖具有较好抑制活性,表现出显著的时间-剂量依赖效应,72h时的IC50值为172.45μg/mL。 OBJECTIVE: To isolate and identify a lipid compound in Haitian melon and determine its anti-tumor activity in vitro. Methods: The crude extract of Haitian melon was extracted with chloroform-methanol-water system (4: 4: 1 by volume). One lipid compound was isolated by continuous reverse-phase column chromatography. The structure of the compound was characterized by IR, ESI-MS, NMR and GC-MS. The effect of the compound on the proliferation of S-180 cells was detected by MTT assay. The results showed that the active lipids were ceramide series compounds mainly composed of 2-amino-1,3-dihydroxy-15-methyl-4-heptadecene long-chain base and 5 kinds of saturated and monounsaturated Fatty acid composition. MTT assay showed that it has a good inhibitory activity on S-180 mouse sarcoma cell proliferation, showing a significant time-dose-dependent effects, IC50 value of 72.45μg / mL at 72h.
