目的 用腺病毒为载体 ,研究p2 7kip1基因转移对食管癌细胞周期的影响。方法 将携带p2 7kip1基因的重组腺病毒转染食管癌细胞株Eca970 6,用四甲基偶氮唑蓝 (MTT)法测定细胞生长抑制效应 ,免疫细胞化学法检测p2 7kip1表达 ,流式细胞仪(FCM)检测细胞周期。结果 p2 7kip1基因转染食管癌细胞株Eca970 6后 ,p2 7kip1表达增强 ,细胞生长显著受抑制 ,GO/G1期细胞比例增加 ,S期、G2 /M期比例降低。结论 腺病毒介导的p2 7kip1基因转移能显著抑制食管癌细胞的生长 ,使细胞产生G1期阻滞。
Objective To study the effect of p2 7kip1 gene transfer on esophageal cancer cell cycle with adenovirus vector. Methods The recombinant adenovirus carrying p2 7kip1 gene was transfected into esophageal cancer cell line Eca9706. The cell growth inhibition was determined by MTT assay. The expression of p2 7kip1 was detected by immunocytochemistry. (FCM) to detect cell cycle. Results The expression of p2 7kip1 was enhanced in esophageal carcinoma cell line Eca9706 after p2 7kip1 gene was transfected. The cell growth was significantly inhibited. The proportion of cells in GO / G1 phase increased and the proportion of S phase and G2 / M phase decreased. Conclusion Adenovirus-mediated p2 7kip1 gene transfer can significantly inhibit the growth of esophageal cancer cells and cause cell arrest in G1 phase.
