目的:研究三七总皂苷对血小板衍生生长因子-BB(PDGF-BB)诱导SD大鼠胸主动脉平滑肌细胞(ASMC)增殖周期的影响及可能作用的机制。方法:将SD大鼠ASMC进行体外培养,采用MTS法检测三七总皂苷对PDGF-BB诱导平滑肌细胞增殖的影响;流式细胞仪分析细胞增殖周期;实时逆转录聚合酶链反应技术检测ASMC中c-fos、cyclin D1和cyclin E1 mRNA表达。结果:PDGF-BB0.02 mg/L能明显升高ASMC光密度值,增加S期细胞比例而降低G0/G1期细胞比例,并上调c-fos、cyclin D1和cyclin E1 mRNA表达。PDGF-BB+三七总皂苷30 mg/L、60 mg/L、180 mg/L组均明显降低PDGF-BB升高的细胞光密度、S期细胞比例而升高G0/G1期细胞比例;明显下调PDGF-BB所致的c-fos、cyclin D1和cyclin E1 mRNA高表达。结论:三七总皂苷可能通过阻止动脉平滑肌细胞由G0/G1期进入S期而抑制PDGF-BB所致的增殖,其作用机制可能与降低c-fos、cyclin D1和cyclin E1 mRNA高表达有关。
Objective: To investigate the effect of Panax Notoginseng saponins on the proliferation of rat aorta smooth muscle cells (ASMC) induced by platelet-derived growth factor-BB (PDGF-BB) and its possible mechanism. Methods: The effects of Panax notoginseng saponins on the proliferation of smooth muscle cells induced by PDGF-BB were detected by MTS assay in vitro. The proliferation of ASMCs was detected by flow cytometry. The mRNA expression level of ASG was detected by real-time reverse transcription-polymerase chain reaction c-fos, cyclin D1 and cyclin E1 mRNA expression. Results: PDGF-BB (0.02 mg / L) significantly increased the optical density of ASMC, increased the percentage of S phase cells and decreased the proportion of cells in G0 / G1 phase and up-regulated the expressions of c-fos, cyclin D1 and cyclin E1 mRNA. The PDGF-BB + Panax notoginseng saponins 30 mg / L, 60 mg / L and 180 mg / L significantly decreased the optical density of PDGF-BB cells and increased the proportion of cells in S phase. The down-regulation of c-fos, cyclin D1 and cyclin E1 mRNA induced by PDGF-BB was highly expressed. Conclusion: Panax notoginseng saponins may inhibit PDGF-BB-induced proliferation by inhibiting the progression from G0 / G1 phase to S phase. The mechanism may be related to decreasing the expression of c-fos, cyclin D1 and cyclin E1 mRNA.
