The effects of captopril on early hemodynamics in diabetic patients and its mechanism were studied by using clearance assay and reverse transcription semi-quantitative polymerase chain reaction (PCR) analysis. Results Captopril could correct the hyperperfusion and hyperfiltration of kidneys in 4-week diabetic rats. Semi-quantitative PCR analysis showed that the non-captopril-treated 4-week diabetic rats had renal, medullary inducible nitric oxide synthase Enzymes (iNOS) gene expression was significantly increased, while captopril treatment of rat kidney, iNOSmRNA medulla was significantly lower than non-treated rats. Conclusions Inhibition of NO production may be one of the mechanisms by which captopril influences the hemodynamics of early diabetic nephropathy.
