目的探讨Periostin(PN)在卵巢癌原位与腹腔转移灶的mRNA表达,研究PN与卵巢癌患者生存期的关系,卵巢癌细胞株SKOV3中PN下调后卵巢癌细胞转移及相关分子机制。方法 Bioconductor软件分析原位灶与腹腔转移灶中PN的表达量。通过KM plot软件分析PN在1 332例卵巢癌患者中的表达量与生存期的关系。通过荧光实时定量PCR检测SKOV3转染siRNA后细胞中Ecadherin和Vimentin的表达量变化,及体外运动能力的变化。结果原位灶中PN mRNA表达量为(6.56±0.27),转移灶中表达量为(9.00±0.19)。PN的mRNA表达水平影响卵巢癌患者生存期,HR值为1.3。转染PN siRNA后,卵巢癌细胞转移能力下降,细胞中Ecadherin表达量提高,Vimentin表达量下降。结论 PN通过调控卵巢癌上皮间质化过程影响卵巢癌的转移能力,在卵巢癌的转移过程中表达量增加,并影响卵巢癌患者的生存期。
Objective To investigate the mRNA expression of Periostin (PN) in ovarian carcinoma in situ and in peritoneal metastasis, and to investigate the relationship between PN and the survival of ovarian cancer patients and the molecular mechanism of ovarian cancer metastasis and metastasis in ovarian cancer cell line SKOV3. Methods Bioconductor software was used to analyze the expression of PN in situ and peritoneal metastases. KM plot software was used to analyze the relationship between PN expression and life span in 1332 ovarian cancer patients. The expression of Ecadherin and Vimentin in SKOV3 transfected SKOV3 cells was detected by real-time fluorescent quantitative PCR, and the change of in vitro exercise capacity. Results The expression of PN mRNA in situ was (6.56 ± 0.27) and that in metastasis was (9.00 ± 0.19). PN mRNA expression levels affect the survival of ovarian cancer patients, HR 1.3. After transfected with PN siRNA, the metastatic capacity of ovarian cancer cells decreased, the expression of Ecadherin increased, and the expression of Vimentin decreased. Conclusion PN regulates the metastasis of ovarian cancer by regulating the process of ovarian epithelial mesenchymal transition and increases the expression of ovarian cancer in the process of metastasis and affects the survival of patients with ovarian cancer.
