目的:观察五子衍宗方对生精障碍模型大鼠睾丸TGF-β1/Smads信号转导通路的影响,探讨其治疗男性不育症的作用机制。方法:采用随机数字表法将50只雄性SD大鼠分为正常对照组、模型组、甲基睾酮组和五子衍宗方低、高剂量组,每组10只。除正常对照组外,其余各组大鼠按醋酸棉酚50mg/kg体质量灌胃给药,制备生精障碍模型。正常组和模型组大鼠灌服蒸馏水,甲基睾酮组大鼠灌服甲基睾酮混悬液,五子衍宗方低、高剂量组大鼠分别给予低、高浓度(含1.75g、3.5g生药/mL)五子衍宗方混悬液灌胃,每天1次,连续30天。光镜下观察大鼠睾丸组织结构变化,免疫组化法检测大鼠睾丸组织TGF-β1、Smad2、Smad4蛋白表达量。结果:棉酚可造成大鼠睾九曲细精管萎缩、退化,管壁变薄,生精细胞减少,排列紊乱,精子发生障碍;睾丸组织TGF-β1表达上调(P<0.01),Smad2、Smad4表达下调(P<0.01)。五子衍宗方能显著下调大鼠睾丸TGF-β1表达(P<0.01),上调Smad2、Smad4表达(P<0.01),拮抗棉酚造成的大鼠睾丸组织结构损伤。结论:通过调控TGF-β1/Smads信号转导通路,改善大鼠睾丸的生精功能,可能五子衍宗方治疗不育症的作用机制之一。
Objective: To observe the effect of Wuzi Yanzong Recipe (TGF) on TGF-β1 / Smads signal transduction pathway of testis in rats with spermatogenic disorder and to explore its mechanism of action in treating male infertility. Methods: Fifty male Sprague-Dawley rats were randomly divided into normal control group, model group, methyltestosterone group and Wuzi Yanzong Fang low and high dose group, 10 rats in each group. Except the normal control group, rats in other groups were intragastrically administered with ginsenoside 50 mg / kg body weight to prepare spermatogenic disorders model. Rats in normal group and model group were drenched with distilled water and rats in methyltestosterone group were given methyltestosterone suspension. Rats in low and high dose Wuzi Yanzong group were given low and high concentration (including 1.75g, 3.5g crude drug / mL) Wuzi Yanzong Fang suspension gavage once a day for 30 days. The structure of testis in rats was observed under light microscope. The expression of TGF-β1, Smad2 and Smad4 protein in testis of rats was detected by immunohistochemistry. Results: Gossypol caused atrophy, degeneration and thinning of the seminiferous tubules, reduced spermatogenic cells, disordered sperm and spermatogenesis in testis, increased expression of TGF-β1 (P <0.01), increased expression of Smad2 Smad4 expression was down-regulated (P <0.01). Wuzi Yanzong Decoction can significantly down-regulate the expression of TGF-β1 in rat testis (P <0.01), up-regulate the expression of Smad2 and Smad4 (P <0.01), and antagonize the injury of testicular tissue in rats. Conclusion: It is possible that Wu Zi Yan Zong Fang can treat infertility through regulating the signal transduction pathway of TGF-β1 / Smads and improving the spermatogenic function in rats.
