目的 运用表达序列标签 (ExpressedSequenceTag ,EST)技术快速、节约、有效地获得有关日本血吸虫 (中国大陆株 )成虫表达基因的知识。方法 随机挑取日本血吸虫 (中国大陆株 )成虫cDNA文库单个重组克隆进行部分测序以获得EST ,获得的EST通过BLAST程序同EMBL寄生虫数据库和GeneBank数据库进行比较及同源性分析。结果 本研究共随机挑取日本血吸虫 (中国大陆株 )成虫cDNA文库单个重组克隆 3 1 4个 ,获得了 1 3 2个有EST价值的序列 ,其中 1 1 3个成功在GeneBankdbEST中登录。 7 6%有EST价值的序列为日本血吸虫已知序列 ,4 5%为日本血吸虫同源序列。曼氏血吸虫或其他生物的同源序列占有EST价值的序列的 2 3 5% ,未知序列占 57 6%。通过同源性分析 ,发现了几个令人感兴趣的基因 ,另外 ,大多数同源序列具有看家基因功能。结论 EST方法具有快速、有效地获得有关日本血吸虫 (中国大陆株 )成虫表达基因的知识的价值
OBJECTIVE: To rapidly, economically and effectively acquire the knowledge about the genes expressed by adult worms of Schistosoma japonicum (Chinese mainland) using the Expressed Sequence Tag (EST) technique. Methods A single recombinant clone of adult Schistosoma japonicum (Chinese mainland) cDNA library was randomly selected to obtain EST. The obtained EST was compared with the EMBL parasite database and GeneBank database by BLAST program and homology analysis. Results A total of 314 randomly selected clones of Schistosoma japonicum (Chinese mainland) adult cDNA library were obtained in this study. One hundred thirty-two EST sequences were obtained, of which 113 were successfully registered in GeneBankdbEST. 7 6% of the EST value of the sequence of the known sequence of Schistosoma japonicum, 45% of Schistosoma japonicum homologous sequences. The homologous sequence of Schistosoma mansoni or other organisms accounted for 23.5% of the EST value of the sequence, and the unknown sequence accounted for 57.6%. By homology analysis, several interesting genes were found. In addition, most homologous sequences have housekeeping gene functions. Conclusion The EST method has the value of quickly and effectively obtaining knowledge about the genes expressed by adult worms of Schistosoma japonicum (Chinese strain)
