目的观察黄芩茎叶总黄酮(Scutellaria baicalensis Stem-leaf Total Flavonoids,SSTF)对Ⅱ型胶原诱导性关节炎(collagen-induced arthritis,CIA)小鼠疾病严重程度和Th17、Treg细胞的影响,初步探讨黄芩茎叶总黄酮对类风湿关节炎(rheumatoid arthritis,RA)的治疗作用。方法选用C57BL/6小鼠,建立鸡Ⅱ型胶原诱导性关节炎小鼠模型,将造模成功的小鼠随机分为CIA模型组、黄芩茎叶总黄酮组、雷公藤多苷组,另设正常对照组于初次免疫后第21天开始灌胃给药,给药35天后,观察各组小鼠关节病理变化,流式细胞术检测小鼠Th17、Treg脾淋巴细胞水平。结果治疗组小鼠的关节损伤减轻,Th17及Treg细胞比例较模型组明显降低(P<0.01)。结论黄芩茎叶总黄酮灌胃给药可以调节CD4+T淋巴细胞亚群Th17/Treg平衡,减轻小鼠关节病理损伤。
Objective To investigate the effects of Scutellaria baicalensis Stem-leaf Total Flavonoids (SSTF) on the severity of Th17 and Treg cells in collagen-induced arthritis (CIA) mice and to explore the effects of Scutellaria baicalensis Stem- Therapeutic effect of stem and leaf flavonoids on rheumatoid arthritis (RA). Methods C57BL / 6 mice were used to establish a mouse model of collagen type II induced arthritis. The mice were randomly divided into CIA model group, total flavonoids of stem and leaves of Scutellaria baicalensis and Tripterygium wilfordii glycosides group, The normal control group began gavage administration on the 21st day after the primary immunization. After the administration for 35 days, the joint pathological changes of the mice in each group were observed. The levels of Th17 and Treg splenic lymphocytes were detected by flow cytometry. Results In the treatment group, the joint damage was alleviated and the ratio of Th17 and Treg cells was significantly lower than that of the model group (P <0.01). Conclusion Scutellaria baicalensis stem-leaf total flavonoid can regulate the balance of Th17 / Treg of CD4 + T lymphocyte subsets and reduce the pathological damage of the joint in mice.
