目的探讨参麦注射液对家兔缺血心肌自律细胞跨膜电位的影响。方法采用细胞内玻璃微电极记录技术描记8只家兔离体心脏窦房结起搏细胞和左心室流出道慢反应自律细胞在正常灌流液(C组)、模拟缺血灌流液(A组)和模拟缺血灌流液+参麦注射液(B组)灌流情况下的跨膜电位曲线,并在模拟缺血灌流20min后测量动作电位幅值、复极50%时间(APD_(50))、复极90%时间(APD_(90))、0期最大除极速率(Vmax)、最大舒张电位、4期自动除极速率(VDD)和自发放电频率(RPF)。结果与C组相比,A组窦房结起搏细胞和左心室流出道自律细胞Vmax、VDD和RPF减少(P<0.05或P<0.01),APD_(50)和APD_(90)增加(P<0.01);与A组相比,B组窦房结起搏细胞和左心室流出道自律细胞VDD和RPF增加(P<0.05或P<0.01),APD_(50)和APD_(90)减少(P<0.05或P<0.01),窦房结起搏细胞Vmax增加(P<0.05)。结论参麦注射液对家兔心肌自律细胞因缺血所导致的电位改变有明显的干预作用,可能是其抗心律失常的机制之一。
Objective To investigate the effect of Shenmai injection on transmembrane potential of ischemic myocardium in rabbits. Methods Intracellular glass microelectrode recording technique was used to characterize the slow response of spontaneous cells of left ventricular outflow tract in the isolated heart of 8 rabbits in normal perfusate group (C group), simulated ischemia perfusion group (group A) And transmembrane potential curve under simulated perfusion perfusion + Shenmai injection (group B). The amplitude of action potential was measured after 20 min of simulated ischemia perfusion, and the APD_ (50) Repolarization 90% of the time (APD 90), 0 maximal depolarization rate (Vmax), maximal diastolic potential, 4 auto depolarization rate (VDD) and spontaneous firing frequency (RPF). Results Compared with group C, the Vmax, VDD and RPF of pacemaker cells and left ventricular outflow tract of atrial node in group A were decreased (P <0.05 or P <0.01), and APD_ (50) and APD_ (90) (P <0.05 or P <0.01), and APD_ (50) and APD_ (90) decreased (P <0.05 or P <0.01) in group B compared with group A. Compared with group A, pacemaker cells and left ventricular outflow tract autonomic cells in group B increased VDD and RPF P <0.05 or P <0.01). The Vmax of pacemaker cells in sinoatrial node increased (P <0.05). Conclusion Shenmai injection on myocardial autonomic cells in rabbits due to ischemia potential changes caused by a significant intervention may be its anti-arrhythmia mechanism.
